فعالیت ضد موتاسیونی سویه‌های پروبیوتیکی جداشده از محصولات پروبیوتیکی |

زمینه و هدف: باکتری‌های پروبیوتیک، مکمل‌های غذایی میکروبی هستند که از طریق حفظ تعادل میکروبی روده تأثیرات سودمندی بر سلامت انسانها دارند. براساس تحقیقات انجام‌شده، استفاده از محصولات پروبیوتیک در کاهش خطر سرطان مؤثر است. هدف از این مطالعه، بررسی فعالیت ضد موتاسیونی باکتری‌های پروبیوتیک جداشده از ماست و قرص پروبیوتیکی بر عوامل جهش‌زا و سرطان‌زا می‌باشد. روش بررسی: در این مطالعه تجربی، باکتری‌های پروبیوتیک از ماست و قرص پروبیوتیکی در محیط MRS در شرایط بی‌هوازی (5% دی‌اکسید کربن و گاز پک) و در دمای C ° 37 جداسازی شدند. سپس با تست‌های بیوشیمیایی شناسایی و مورد تأیید قرار گرفتند. در ادامه، اثر ضد موتاسیونی سوپرناتانت کشت آنها در مقابل عوامل موتاسیون‌زای سدیم آزید و پرمنگنات پتاسیم توسط تست ایمز (سویه سالمونلا تیفی موریوم TA100 ) در حضور و عدم حضور S 9 ارزیابی شد. یافته‌ها: در این مطالعه از ماست و قرص پروبیوتیکی، 6 باکتری پروبیوتیک جدا شد. نتایج فعالیت ضد موتاسیونی باکتری‌ها توسط تست ایمز نشان داد آنها می‌توانند عوامل موتاسیون‌زا را در برخی گونه‌ها تا بیش از 40% مهار کنند که فعالیت ضد موتاسیونی خوبی محسوب می‌شود. نتیجه‌گیری: نتایج این مطالعه نشان داد استفاده از محصولات حاوی باکتری‌های پروبیوتیک، دارای اثرات ضد موتاسیونی و ضد سرطانی بسیار خوبی می‌باشد. آنها با تغییر فلور روده سبب کاهش جذب مواد سرطان‌زا و جهش‌زا شده و به سلامت انسان کمک می‌کنند. |

Evaluating Antimutagenic Activity of Probiotic Bacteria Isolated from Probiotic Products |

Background and Objectives: Probiotic bacteria are microbial nutrition supplements which have useful effects on human health by maintaining of bowel microbial balance. There are many studies that have been suggested the use of probiotic products as cancer risk reducer. The aim of this study, is isolation and detection of probiotic agents from yoghurt and probiotical tablet and evaluation of their abilities to decrease some effects of mutagenic and carcinogenic agents. Methods: In this study, probiotic bacteria were isolated from yogurt and probiotic tablet by using MRS in anaerobic condition (5% Co 2 and gas peck) and temperature of 37°c. Then, they were detected by using biochemical tests. Their anti mutagenic effects of supernatant culture were evaluated against mutagenic agents of azid Sodium and Potassium Permanganate by ames test ( Salmonella typhimurium TA100 ) in presence and absence of S 9 . Results: Six probiotic bacteria were isolated from yogurt and probiotic tablet. Their anti mutagenic activity results based on ames test showed they can inhibit mutagenic agents more than 40% in some species, which is considered as a good result. Conclusion: The results of this study show that the use of probiotic bacteria found in different products such as yogurt and probiotic tablets, have proper anti mutagenic and anti carcinogenic effects. They change the micro flora of bowel and, as a result, reduce absorption of mutagenic and carcinogenic agents and help to maintain human health. |
