عنوان فارسی فراوانی چند شکلی آلل rs۸۰۹۹۹۱۷ مربوط به ژن اینترفرون لامبدا-۳(IL-۲۸B) در جمعیت ایرانی
چکیده فارسی مقاله زمینه و هدف: (IL-28B)IFN-&lambda3 یکی از سیتوکاین‎های جدید و عضوی از خانواده اینترفرون‎های تیپ III است. فعالیت‎های بیولوژیکی متعددی از آن در تنظیم سیستم ایمنی، عملکردها در تومور و متاستاز و یا نقش در بیماری‎های ویروسی از جمله هپاتیت B و C نشان داده شده است. در این مطالعه به بررسی میزان فراوانی پلی مرفیسم آلل rs8099917 در ژن IFN-&lambda3 جمعیت ایرانی می‎پردازیم. مواد و روش‎ها: در این مطالعه تجربی- آزمایشگاهی، پس از خون‎گیری از 118 نفر از افراد ایرانی غیر خویشاوند، استخراج DNA مطابق دستورالعمل کیت انجام شد. برای بررسی پلی مورفیسم آلل rs8099917 مربوط به ژن IFN-&lambda3 از تکنیک Nested-PCR و روش RFLP با آنزیم‎های محدود الاثر BsrDI و Tsp45I استفاده گردید. یافته‎ها: پس از انجام آزمون‎ها و جمع‎آوری داده‎ها فراوانی واریانت‎ها به صورت ژنوتیپ GG، 5/2 درصد، ژنوتیپ GT، 4/31 درصد و ژنوتیپ TT، 1/66 درصد بود. آلل G، 22/18 درصد افراد و آلل T، 78/81 درصد افراد را در برمی‎گرفت. نتیجه‎گیری: نتایج این مطالعه نشان می‎دهد که ژنوتیپ TT و آلل T فراوانی غالب در پلی مورفیسم آلل rs8099917 در ژن IFN-&lambda3 در جمعیت ایرانی می‎باشد. با توجه به عملکردهای مختلفی که برای IFN-&lambda3 نشان داده شده است، مشخص شدن فراوانی این واریانت می‎تواند راهگشا و کمک کننده به بسیاری از مطالعات، طراحی و بررسی‎ها و هم‎چنین راهکارهای درمانی در افراد ایرانی در بیماری‎هایی نظیر تومور و هپاتیت باشد.

عنوان انگلیسی Distribution of Rs8099917 IFN-λ3 (IL-28B) Allele Polymorphism in Iranian Population
چکیده انگلیسی مقاله Background: IFN-&lambda3 (IL-28B) is a new cytokine from the type III interferons. Its role in several biologic activities, such as regulation of immune responses, tumor and metastasis, or its functions in HBV and HCV diseases, has been shown. The present study evaluated the frequency of rs8099917 allele polymorphism in IFN-&lambda3 gene in Iranian population. Materials and Methods: In this laboratory-experimental study, after collecting blood samples from 118 unrelated Iranian subjects, DNA extraction was performed according to the kit protocols. Evaluation of rs8099917 allele polymorphism was done by Nested-PCR and RFLP method using BsrDI and Tsp451 restriction enzymes. Results: After running the tests and data analysis, the frequencies of GG genotype (2.5%), GT genotype (31.4%), and TT genotype (66.1%) were obtained. Allele frequencies were 71.22% for G allele and 81.78% for T allele. Conclusion: The results of this study showed that TT genotype and T allele are the dominant frequencies of rs8099917 allele of IFN-&lambda3 gene in Iranian population. Regarding the different established functions of IFN-&lambda3, frequency determination of this variant can be helpful in different studies, designs, evaluations, and treatment procedures in some diseases, such as tumors and hepatitis, in Iranian patients.
