اثر عصاره متانولی بذر خارمریم بر میزان گلوکز، شاخص‌های اکسایشی و فاکتورهای بیوشیمیایی در سرم موش‌های صحرایی دیابتی نر ویستار |

اهداف: دیابت یک اختلال متابولیک است که منجر به افزایش استرس اکسیداتیو و کاهش سیستم دفاع آنتی‌اکسیدانی می‌شود. هدف مطالعه حاضر، تعیین اثرات عصاره متانولی بذر خارمریم بر سطح گلوکز، شاخص‌های اکسیداتیو و فاکتورهای بیوشیمیایی در موش‌های صحرایی دیابتی بود. مواد و روش‌ها: در این مطالعه تجربی، 24 سر موش صحرایی نر سفید نژاد ویستار به‌طور تصادفی به چهار گروه 6تایی؛ کنترل سالم (کنترل‌منفی)، کنترل دیابتی بدون تیمار با عصاره (کنترل‌مثبت) و دو گروه دیابتی تحت تیمار با دو دوز 150 و 100میلی‌گرم بر کیلوگرم وزن بدن عصاره متانولی تقسیم شدند. القای دیابت با استفاده از داروی استرپتوزوتوسین به‌صورت تک‌دوز و داخل‌صفاقی به‌میزان 60میلی‌گرم بر کیلوگرم صورت گرفت و پس از مشاهده علایم دیابت تیماره عصاره‌ها به‌مدت 4 هفته به‌صورت روزانه و داخل‌صفاقی انجام شد. داده‌ها با استفاده از آزمون آماری تحلیل واریانس یک‌طرفه و آزمون اندازه‌گیری مکرر مورد تجزیه و تحلیل قرار گرفتند. یافته‌ها: تیمار با عصاره متانولی بذر خارمریم سبب کاهش وزن، سطح گلوکز خون، کلسترول، مالون‌دی‌آلدئید و پروتئین کربونیل و افزایش میزان HDL و فعالیت آنزیم پاراکسوناز در گروه‌های تیمار نسبت به گروه کنترل دیابتی شد که موثرترین غلظت عصاره در کاهش کلسترول، گلوکز، پروتئین کربونیل، مالون‌دی‌آلدئید و افزایش فعالیت آنزیم پاراکسوناز 150میلی‌گرم بر کیلوگرم و موثرترین غلظت در افزایش HDL و وزن 100میلی‌گرم بر کیلوگرم بود (05/0p<). نتیجه‌گیری: تزریق عصاره متانولی بذر گیاه خارمریم سبب ایجاد تغییرات مثبت در شاخص‌های اکسیداتیو و فاکتورهای بیوشیمیایی در موش‌های صحرایی دیابتی می‌شود. |

Effect of Methanolic Extract of Silybum marianum Seed on the Levels of Glucose, Oxidative Indicators and Biochemical Factors in the Serum of Male Diabetic Wistar Rats |

Aims: As a metabolic disorder, the diabetes increases the oxidative stress, while it reduces the anti-oxidant defense system. The aim of the study was to determine the effects of the methanol extract of Silybum marianum on the glucose level, the oxidative indices, and the biochemical factors in the diabetic rats. Materials & Methods: In the experimental study, 24 male and white Wistar rats were studied. The rats were randomly divided into four 6-rat groups including healthy control (negative control), diabetic control without any extract treatment (positive control), and two diabetic groups treating by 150 and 100mg/kg methanol extract respectively. The diabetes was induced by 60mg/kg one-dose streptozotocin as intra-peritoneal injection. The diabetes symptoms having been observed, 4-week and daily extract treatment was done as intra-peritoneal injection. Data was analyzed using one-way ANOVA and the repeated measurement tests. Findings: The treatment, done by the methanol extract of the seeds of Silybum marianum, reduced weight, the blood glucose level, cholesterol, malondialdehyde, and protein carbonyl, while it increased HDL and the activity level of paraoxonase enzyme, in the treatment groups compared to diabetic control group. In addition, the most effective extract concentration, reducing cholesterol, glucose, protein carbonyl, malondialdehyde and increasing the activity level of paraoxonase enzyme, was 150mg/kg. And the most effective concentration, increasing HDL and weight, was 100mg/kg (p< 0.05). Conclusion: The injection of methanol extract of the seeds of Silybum marianum positively changes the oxidative indices, as well as the biochemical factors, in the diabetic rats. |
